An increasing quantity of evidence supports a force-driven mode of activation of the Notch signalling pathway, whereby exposure of the critical S2 site in the Notch NRR occurs upon mechanical unfolding of the protein during transendocytosis of the Notch extracellular domain into the ligand presenting cell.
